In collaboration with Prof. Yang Qu (UNB), we identify and characterize three new cytochrome P450 (CYP) enzymes that facilitate regio- and stereospecific oxidation of four monoterpene alkaloid (MIA) skeletons: iboga, aspidosperma, pseudoaspidosperma, and quebrachamine. From CYP genes identified by molecular biology and phylogenetic analyses, 3D models of the CYP enzymes were derived using the AlphaFold 3 AI modeling facility.
Docking various MIA substrates to the CYP models revealed substrate selectivity and exact location of the substrate oxidation sites.
